Subtle active site differences have large effects

Subtle differences in active site composition can often yield quite startling differences in enzymatic activity.  In a recent collaborative paper between Colin Jackon's lab and ours, we show that changing the hydrophobicity of catalytically relevant regions of the active site can make a large difference to reactivity towards some substrates in natural variants of the triazine hydrolase, TrzN.
